Several studies in man and experimental animals have clearly documented the progressive accumulation of aminoglycosides in the prilymph and enolymph in the inner ear. Gentamicin is a nephrotoxic agent that caused acute renal failure in experimental animals. The specificity of its toxicity is related to its accumulation in the renal proximal convoluted tubules, enhancing the production of hydrogen peroxide. Allopurinol (Allo.) is an inhibitor of xanthine oxidase and prevent the conversion of hypoxanthine and xanthine to uric acid and subsequent free radical that primarely cause tissue injury. Inhibition of this enzyme accounts for the major pharmacological effects of Allo. It’s treatment increases the levels of glutathione peroxidase in the kidney tissues suggesting its beneficial effects against renal injured rats.
